Auto news on Youtube Sep 29 2017

Hello there.

You know, I get this question all the time – both from my students in my classes and

on YouTube – what is the difference between 'can' and 'could'?

Well, in this lesson, I will teach you the three main differences between these modal

verbs and I'll show you how to use them correctly without making mistakes.

As always, there is a quiz at the end of the video to test your understanding.

The very first thing you should know is the basic use of 'can'.

This verb is used to show ability.

Here are some examples: I can play the guitar.

He can lift heavy weights.

She can beat anyone at chess!

So, all of these express ability.

What about what 'could' then?

Well, 'could' is just the past tense of can.

I can say: I could play the guitar when I was in school.

That means, I had the ability in the past, but now I can't play the guitar because

it's been a long time and I'm out of practice.

Here a couple more examples: He could lift heavy weights until he had the accident.

She could beat anyone at chess in her prime!

'In her prime' means in her best years.

Maybe when she was younger and was playing competitively.

So in these sentences – 'can' shows present ability, and 'could' shows past

ability.

This is the first difference between the two words.

The second difference is when we want to talk about possibility.

To talk about what is possible, we normally use 'could' and not 'can'.

Now, imagine that you and me, we're both waiting for a train at a train station.

It's time for the train to arrive but we don't see it.

So I might say: Hmm, the train could be late.

So I'm making a guess about the current (or the present) situation, saying what is

possible.

Here's another one: If you don't study, you could fail the exam.

Are you sure to fail the exam?

No.

You could pass.

But I'm saying that it's possible that if you don't study, you'll fail.

Notice that this sentence talks about future possibility.

Ah, but I have a question – we just talked about present and future possibility.

Can we talk about past possibility?

Yes, we can.

Here's an example: Robert could have gotten the job if he had gone to the interview.

So what do we understand from that?

Well, there was a job interview.

It was possible for Robert to go to the interview and get the job.

But he didn't go.

So he didn't get the job.

Now, as I said before, we use 'could' in all these sentences to talk about what's

possible in the past, present, and future, but there is one situation where you can use

'can' and that is when you want to talk about general possibility.

For example: You can catch a cold if you go out in the rain.

Traveling across Europe can be quite expensive.

You can see that these are just general statements about what is possible in the world.

In this case, it's OK to use 'can', but this use is less common.

Normally, when we talk about possibility in our lives we use 'could'.

Remember that.

Alright, let's now move on to the final use of 'can' and 'could', and this

is in polite expressions when we talk to people.

There are four situations you need to know about.

The first is making suggestions.

For this purpose, we use 'could'.

Here are some examples: We could try that new Italian restaurant tonight.

You could ask your sister to lend you some money.

In the first sentence, I'm making a suggestion about where we can have dinner.

In the second, I'm suggesting that if you need money, why don't you ask your sister?

Using 'could' just makes it very polite.

The second situation is making requests.

Here, both 'can' and 'could' are possible: Can you pass me the salt?

Could you pass me the salt, please?

Or on the phone: Can I speak to Rahul?

Could I speak to Rahul, please?

Did you notice that I said 'please' at the end of the sentences with 'could'?

That's because 'could' is more polite than 'can'.

So if you are in a formal situation, like if you're talking to your boss or other

superior and you want to be very polite, use 'could'.

If you're with friends or family, then 'can' is fine.

Our next function is making offers: Can I get you something to drink?

Could I get you something to drink, sir?

Can I help you with that?

Could I help you with that, madam?

Again, notice the extra politeness with 'could'.

Finally, the last conversational situation is when we want to ask for permission.

Here are some sentences: Can I borrow your pen?

Could I borrow your pen, please?

Can I take the day off tomorrow?

Could I take the day off tomorrow?

If you're very friendly with your boss at work, you would use 'can', but if your

boss is very strict, then you would use 'could'.

OK, if you're ready, it's now time for the quiz to see if you can use 'can' and

'could' correctly.

On the screen, there are eight sentences.

In each one, I want you to fill in the gap with 'can' or 'could'.

Stop the video, think about your answers, then play the video again and check.

OK, here are the answers.

How many did you get right?

in japanese, the phrases are already chosen

i guess it kind of shows

how much customer service means to them

and so in the case of american fast food...

the one and only... is obviously mcdonalds

and honestl american and japanese mcdoanlds are pretty different

so today i want to look at american mcdonalds

and the japanese mcdonalds

What's the Difference Between Affiliate Marketing and CPA Marketing - Duration: 3:44.

hey guys it's Brian from BG media innovation and I got a great one for you

here today so I got this question from the last video that I posted which was

you know basically what the best a business opportunity behind Amazon

private labeling was and my answer to that was basically affiliate marketing

or cpa marketing now they're relatively similar but they're slightly different

and i got that question you know what's the difference between affiliate

marketing and cpa marketing so i'm gonna here to answer that for you basically in

a couple short sentences i'll keep this one short for you today but if you

didn't know obviously or you didn't see that last video check that out i'll link

that in the description for you below that's a very good one basically goes

into the suit the summary of you know the courses that i have out which email

an affiliate marketing mastermind it's a great one

basically this deals with email marketing and affiliate marketing and

how they combined and how you can combine them to make a lot of money and

cpa marketing mastery and innovation is one that i just recently actually put

out fairly recently and is doing really well and that basically deals with just

cpa offers and how you can apply them so the difference between affiliate

marketing and CPM marketing and this is not necessarily well first and foremost

if the difference between affiliate marketing and cpa marketing is affiliate

marketing is you know you're promoting a software or a service or whatever the

case may be and you're an affiliate of that so you if someone purchases through

your link you would get the affiliate commission on that whatever that's

deemed now there are a bunch of affiliate networks out there i'm not

gonna go into these things you know in-depth because i have entire courses

on them so you can check the courses out to they'll be in the the links for them

will be in the description too but the difference where cpa marketing differs

from affiliate marketing is obviously you're still getting people to sign up

through your links and stuff like that but you're not they're not signing up

and paying for a service per se now that that could be the case sometimes in cpa

marketing but for the most part the way I teach cpa marketing is to go after the

free lead ones so you know you're gonna get a higher customer acquisition

basically on cpa because the majority of the of the things that you're promoting

our free offers so someone signs up basically through with their email

through your link you are the difference is nobody's purchasing you're just

acquiring customers for the most part that's the way i teach it at least

so you're obviously gonna get a lower customer acquisition on on affiliate

marketing because people are paying for the service through your direct link and

if you're familiar with any of my courses obviously you know I promote

affiliate offers on the back end of all my courses things that are related to

that course and there's always obviously always affiliate offers in the YouTube

comments or in the YouTube comments the YouTube description sorry but cpa

marketing is different because you're basically promoting something free so if

a company was paying you you know to acquire customers emails you would

basically promote their cpa offer and when people click your link and they

sign up with their email the company would pay you because they're a

potential leads so that's the slight difference at least in the way that i

teach it obviously you're going to get a lot more people and commissions through

cpa marketing but the Commission's will be a lot lower so you get higher

commission or higher rates but uh i mean its are higher amount of people signing

up but lower commissions whereas affiliate marketing slightly you're

gonna get a lot lower people but higher commissions so it's a little bit

different it differs slightly in the way they promote them not a whole lot but

like i said both these courses are great if you're interested in them please

check them out they'll be in the description box for you below i'll also

link the other like i said the other from the previous video in the

description below if you want to check that out so i hope that you guys like

this video if you liked it please like the video comment and subscribe but it

really helps the channel grow and i'll see you guys in the next one

What's the Difference Between qPCR vs Qubit for Library Quantitation? - Duration: 4:34.

Hi Everyone, my name is Efi Melista and I'm the Head of Field and Lab Operations at Omixon.

And I tend to go to a lot of labs to train people on NGS for HLA typing, using Holotype HLA,

and very often one of the questions that a lot of people ask is what is the difference

between qPCR versus Qubit for library quantitation?

So I figured we should create a little video to discuss all of this.

So today we will be talking about should we do qPCR for library quantitation, or should

we do Qubit for library quantitation?

What is better?

What is the difference?

What should you use?

The straight answer is that they're both the same.

Well… they work differently, but essentially they give you the same kind of information.

They both measure the concentration of your library and help you guide how much you dilute

to load on the MiSeq.

The important thing to remember is that you always want to make sure that the concentration

that you're measuring of your library is accurate enough in order to do an accurate

dilution because if you overload the MiSeq, it will end up in over-clustering; if you

underload the MiSeq, it will end up in low cluster density, which means that you will

not generate enough data for all of the samples and loci that you are running.

So it is very important to have an accurate measurement.

So what's the difference?

The qPCR is able to quantify the library using Illumina-specific primers.

The kit rather, that we use, the Kapa Biosystems kit, it uses Illumina-specific primers that

are complimentary to the Illumina-specific adaptors, that are already ligated on the

indexed libraries.

So this way it is able to measure how much is the adaptor-ligated library – how much

is the adaptor-ligated material that you have in your sample.

So it's a very-very accurate measurement, so when you dilute that down, to a certain

concentration, you know that you're diluting a very accurate measurement.

So what's the difference with the Qubit then?

The Qubit on the other hand, it uses a double-stranded DNA fluorescent dye, which is pretty much

an equivalent to a SYBRgreen dye.

What this means is that it intercalates to anything double-stranded in your sample.

So it is able to measure anything that is double-stranded in your library.

Is this an accurate measurement?

It's not as accurate and specific, as the qPCR.

But is that an incorrect measurement?

Actually it's not.

And the reason why is because when you enter that value into the excel workbook that we

provide with the Holotype HLA kit, then you can actually calculate very accurately what

is the true amount of your library in your sample.

In the workbook, for this calculation, we actually use a standard curve in the background

that has been created with more than a hundred different libraries that have been quantified

both with qPCR and Qubit.

So that helps remove any variability in this calculation.

So it's very convenient that way.

So you're going to tell me "which one should I use"?

Well, you can use either one: the Qubit will help you save time, the qPCR, however, is

a bit more specific.

The qPCR, I would say it is more informative when you're in a troubleshooting mode.

So if you have figured out that something went wrong into the process, into your whole

workflow, the qPCR can actually help you give you more information to tell you where did

things go wrong in a way.

So it's a bit more informative.

But for good, high-quality libraries, either one will do.

So it's up to you, the choice is yours!

If you want to know more about it, feel free to contact us at support@omixon.com or make

sure you visit our website, omixon.com and the MyHolotype registration area, where you

can actually find a lot more information.

Thank you for watching!

Bye!

Education Made a Difference - Michael Collier | Ashford University - Duration: 0:35.

It was about a month and a half before graduation.

I got a LinkedIn message from a former colleague that I worked with, and she let me know, 'hey,

there's a job that I think would be perfect for you.'

So I looked into it, and the job happened to require a bachelor's degree.

Well, since I was in my last class, they were willing to bring me on.

My salary from the time I left to when I came back doubled, so I can only say that the education

makes a huge difference, and it opened doors that would have been shut.
