The musical does not let down. It gets all the gems, all the one-liners from the

movie, are put in there, you know, for the fans, but then, there's a couple twists

you wouldn't expect, a couple little new rewrites and interesting storyline

changes that'll keep people intrigued and entertained. I think that they're gonna

love it.

Well, it's cool to see, like, they're gonna be like, "what's a pager" and

like, "What's a flip phone, what is that?" so it's good to get, like, what we went

through growing up.

I think that everybody can kind of fall in love with

it in a new way. If you are a huge die-hard fan of the original, we have

all of the classics, we got your favorite lines, we have your favorite characters,

we're hitting the costumes, and then we're also putting our own twist on it.

Everyone's version of their character, no one's going for the original way that

it was presented. Everyone's able to put their own spin on it and I think that new

people that haven't seen this before are gonna have to go back and

see the original, and I think it'll be a nice melding of the two.

I definitely

think that it's funny and energetic and that people who have seen the movie will

definitely love it, and anybody who lived through the 90s is gonna like, lose their

minds because it is such a great reminiscing experience.

There's a

beautiful heart to this show that I think people are gonna really fall in

love with. There's a reason why this movie has lasted for so long, and the

musical absolutely carries on that tradition and brings it to a new

audience, a younger audience, fresh ears, fresh eyes and the music that we're

doing... I mean, I get to sing some of the greatest, like, 90s hits that I would sing

in the car when I was a kid, you know, driving down with my friends, and to get

to sing those on stage... it's a dream come true, it's amazing.

I think we do a

really good job of, like, satisfying both. Because you don't have to have seen the

movie, you don't have to come in with any point of context other than wanting to

have a good night of theater. And you will have a good night of theater but if

you want to see Clueless on stage too, you'll have that. We got both, you know?

I think that we have done, so far, a really good job putting what the

longtime fans of Clueless the movie, I feel like we've translated that really

well to the stage, so I think that they're gonna be really excited. I hadn't

even thought about the fact that this will be some people's first experience

with Clueless, and I think that, not only are they going to love this story, but

it's just gonna make them want to go home and watch the movie.

I think if you

love the movie, you'll, like, 100% be obsessed with the musical. Amy Heckerling

is involved in the process, so it is the movie, like, on stage, which I feel like is

fun, and it's a 90s throwback musical, which everybody can, like, love and enjoy,

It hints at the original, but it's also very new, and I think the 90s songs that

are in it will also, I feel like they're gonna blow their minds with like, you

know, when you hear that song you're like, "Oh, no way! That song is in the show?" and

I think that's gonna really win over the crowd.

If they've come to the show

already being a fan of Clueless, they're gonna get a next level of enjoyment and

understanding of this movie that I'm sure they've grown up knowing and loving.

So you are getting at just one arm?

Both of my arms.

Oh wow.

For most patients with acute lymphoblastic leukemia, the kind of cancer Bo Cooper suffers

from, standard treatment can offer a cure.

But Bo is among a very small group whose leukemia has proven incurable.

Back in Canada, Bo received chemotherapy, standard initial treatment, for leukemia.

He then received a bone marrow transplant

and none of those treatments have been able

to keep the leukemia away.

Hey Doc.

How are you doing?

Good.

Yeah?

I'd shake your hand, but there is a needle in there.

You're attached.

It's kinda hard to move.

With all standard therapies having failed Bo, Dr Fry and his team will be trying something

radical - re-engineering Bo's own immune system to cure his cancer.

The first step in that treatment is for Bo's T cells, a type of immune cell, to be removed

from his blood in a process called Apheresis.

Cells get separated in the machine into the different components.

The part that we need goes into a bag, the rest of it goes back into you in the other arm.

Does that make sense?

Yeah.

Sounds good.

Each leukemia cell in Bo's body is marked with different proteins on its surface.

Among them, one that is called CD22.

Since Bo's immune system doesn't recognize his leukemia is a threat, Dr Fry will genetically

modify one part of it, his T cells, so that they can recognize and attach themselves to

the CD22 protein and kill his cancer.

This modified T-Cell is called a Chimeric Antigen Receptor T-Cell, or CAR T-Cell for short.

It is at the cutting edge of cancer research and Bo will be the first leukemia patient

ever to receive the highest dose of these immune cells.

There's really no other option.

Without this working, Bo doesn't survive.

If this does not work for Bo, he's run out of time.

Finding a cure for cancer patients like Bo Cooper, who do not respond to standard treatment

is the holy grail of medical science and has proven to be one of the most difficult and

complicated questions facing humanity.

Nearly 40 years ago, right here at Building 10, one man began a personal quest for this

elusive cure.

Miraculously, he may have found the answer lurking within ourselves.

My voyage in this adventure, if you will, began when I was a junior resident in surgery

and a patient came in, a veteran in his 60s, with right upper quadrant pain.

It seemed to be a typical gall bladder attack.

But as I went through his chart, an astonishing story evolved.

12 years earlier, he had been operated on for a cancer that had started in his stomach.

The surgeons described it having spread to the liver and the patient's abdomen was closed,

nobody expected him to survive.

But as I flipped the pages of this chart, he was back in three months feeling okay.

Six months later he was gaining weight.

A year later, he was back at work.

And here he was 12 years later completely cancer free.

What Dr Rosenberg witnessed seemed impossible.

A person who cured himself of cancer.

Some how that patient's body had learned to destroy that cancer, had recognized it as

foreign, eliminated it just they way it eliminates a virus from a common cold.

It got me thinking that there must be something in that patient's body that caused the regression

and in fact the immune system is our major defense.

The idea that our own immune system is capable of fighting cancer was so radical that when

Dr Rosenberg began his research there wasn't even a name for this experimental treatment.

The main problem with your immune system recognizing cancer is that cancer, for the most part,

it really is self.

It's self gone bad.

So how do you tell your immune system that it's bad?

That's, that's the tough part.

For a decade, Dr Rosenberg tried to train the immune system to fight cancer in a number

of first In Human trials.

For years he failed.

But in 1985, he successfully cured his first patient with a treatment that has since become

known as Immunotherapy.

Activating the immune system for cancer treatment, so called Immunotherapy, is the most exciting

science in the whole cancer field because it's really working.

Today, Immunotherapy is the basis for many of the trials at Building 10, including the

one Bo Cooper is enrolled in.

You want to eat?

No.

You hardly ate anything.

In a Building 10 lab, Bo's immune cells are being genetically re-engineered in the hope

that they can recognize and fight his cancer.

Car T-Cells, that's what we're here for.

That's what we want.

Waiting is the hardest part.

Bo is scheduled to receive 200 million of these genetically modified immune cells.

But the work to create them in the lab is painstaking and can be unpredictable.

It's important for us to have a sense that, that process of making those Car T-Cells is

going to be successful.

I need to see as high of a number as they can possibly achieve.

The more cells the better.

What's the culture look like numbers wise?

This is today's?

Yeah, these are today.

Yeah, it dropped.

So you're read now at about 13% which isn't great.

The number of those cells is lower than I would have hoped for.

You know, so he may or may not need target dose, you know, but I mean, again, none of

this is certain until we see the final product next week, so...

We really can't wait before we treat him.

He clearly has progressive leukemia.

It's still early, it's gonna be low.

Yeah.

I mean there's something there.

The number of Car T-Cells that I have is not enough.

Dealing with these sorts of trials, it's a roller-coaster ride.

What's that sound?

Do you know what that sound was?

Here's another video.

That was a plane creating a shock and breaking the sound barrier.

Recently, there have been a few of videos by Destin on smartereveryday where he talks

about bullets and whips producing shocks.

But what exactly is a shock?

Let's find out on today's episode.

Hi I'm Nikhilesh and I'm Kushal and we are the 2BrokeScientists

At the molecular level, fluid particles travel in a continuum.

When an obstruction is suddenly introduced into the flow, something interesting happens.

The particles in front of it slow down instantaneously, creating an increase in local pressure.

Since the fluid is a continuum, the information of whatever happens in one place is transmitted

to other regions in the form of pressure waves.

This pressure wave then causes the oncoming fluid to adjust its properties.

Don't these pressure waves remind you of something?

Doesn't sound also travel in the form of compressions and rarefactions through a medium?

The information propagation in a fluid happens through acoustic waves which travel at the

speed of sound.

What if the fluid particles are traveling faster than the speed of sound?.

Since the information transfer cannot happen faster than the speed of sound, these fast-moving

particles don't have any information about the obstruction in front until they actually

reach it.

The moment they reach it, an extreme collision of particles with themselves and the object

occurs which manifests itself as a shockwave.

A good analogy to information transfer in fluids are traffic waves.

Take a look at this busy roundabout where one of the cars suddenly slows down.

The driver behind this car receives information in the form of visual signals and he adjusts

his speed.

You can clearly see this wave of information passing backward even though the cars are

still moving forward.

If the cars were moving so fast that the drivers were not able to process the visual information,

they would then crash into each other.

That's exactly what happens in a shock wave.

You would have heard of the Mach number, a dimensionless number is very important while

studying shocks.

Mach number is basically the ratio of the velocity of a fluid to the velocity of sound

in that medium.

So clearly, if the Mach number is greater than 1, it means the fluid travels faster

than the speed of sound and there's no time for information to propagate.

These are called supersonic flows.

A moving shock is called a shock wave which is very thin somewhere in the order of 200

nm and in this region, the fluid undergoes sudden changes in properties.

The Mach number decreases across a shock wave along with a sudden rise in pressure, density

and temperature.

A shock wave is also an irreversible process because of the extreme reactions produced

by the collisions of the particles.

In some applications of aerospace engineering, we try to avoid shocks because they cause

vibrations and an increase in drag.

Even in commercial aircraft, while the plane itself might not go beyond M1, there are regions

over the wing where the flow can locally become supersonic.

This is the exact reason why most commercial planes have their wings swept back.

When air now flows over the wing, it sees a thinner wing because only a component of

the velocity is perpendicular to the wing and now does not accelerate to supersonic

speeds.

When a stationary object emits sound waves at a certain frequency, the waves are generally

spherical in nature and travel at the speed of sound.

The moment an object starts moving, the sound waves in front of the object compress increasing

the apparent frequency and the ones behind elongate decreasing the apparent frequency.

This is called the Doppler effect.

When the object travels faster than the speed of sound, the sound waves that are emitted

form a cone known as the Mach cone The thunderous clap that you heard at the beginning of the

video was when the camera enters the shock cone.

This phenomenon is what engineers took advantage of while designing stealth aircraft like the

SR 71 Blackbird, which travels at supersonic speeds.

By the time people on the ground heard the plane, it was well beyond the reach of missiles.

Shock waves produce a lot of changes in the properties of a fluid, but it's not always

easy to visualize change like pressure or density with our eyes.

That's where interesting visualization techniques like schlieren and shadowgraphy become useful.

Mediums with different densities have different refractive indices meaning that if light passes

through the medium, it changes direction.

We use this property of light to see all the cool images of shock waves.

So shockwaves aren't so shocking after all!!

I decided to film a Finnish vlog since I'm here in Finland

It's so beautiful because it's been humid and cold

The trees are covered in ice

I wish the sun would shine, then it would be perfect photo weather

But this will be ok

I hope it looks the same at Hyvinkää

You never know about the weather, but it looks promising right now

I'll take Snö with me, he'll be my assistant

I drove the wrong way twice

We got lost with Snö

But now we're finally here

I'll go looking for the barn door

And my dog is impatient because we've been driving around

He doesn't understand Finnish

Now we're on the photoshoot trip

The surroundings are so beautiful

from the heaviest I was was 350 I got all the way down to a nice and clean one

eighty five nine percent body fat and have gone and kind of stayed around that

direction my whole entire life some things that are going to be

important that we're going to talk about in this channel other than just fitness

journeys its debunking myths I hate snake oil I hate people that say

one thing promote a product and it's really not doing what it does you're

gonna see a whole lot of that in the fitness industry people that I'll tell

you they are using CBD oil they'll tell you they are using this supplement that

supplement to get their gains when they are totally using something else and

it's a lie we're going to use science-based information to let you

guys know what really works what doesn't what's achievable for someone who's

natural versus someone who's used in performance enhancers we're going to

started we need a new one for this week and mm-hmm

question now this item has to be an actual existing item not something like

a teleporting device or something you can't just make something up it can't be

something that's theoretical or non-existent has to be something that

actually exists so what item would make your life much easier right now leave

of quality of the video I have this now set to 60fps but the problem is the

resolution has to of the camera has to be at 720 for that to work instead of

watching 1080p for me to have 60fps camera resolution has to be 720p so 720

blown up to 1080 let me know what you think about that it might look a little

life I want to forget my old life and open a new page in my life starting from

gotta change it I guess not too much else to really say regarding that just

you can do it though and that is it and for my new year's resolution or whatever

I don't I know I don't really care too much about the the resolution stuff

either I just I guess I said resolution because that's kind of the resolutions

are at the traditional thing so but my goal for this year is a couple things

one to read a little more because last year I the beginning of the year

actually I was reading a bit not a lot but a bit and I stopped halfway through

or something for some reason and part two is to be a little bit less lazy and

to catch up on things and I'm Way behind on so yeah we'll see how well that goes
